When a virus, bacterium or other antigen enters the body, B cells are activated and secrete large numbers of antibodies that recognize and bind to the antigen to mark it for destruction or neutralization. Unfortunately, a subset of these antibodies may also bind to conserved domains in other proteins. This is why it’s important to avoid peptide sequences that are highly similar to conserved protein structures and to favor those that are more flexible and solvent-exposed.

In the 1970s, Kohler and Milstein discovered a way to generate large quantities of antibodies with predefined specificity (monoclonal antibodies), which significantly enhanced the usefulness of these antibodies. Monoclonal antibodies exhibit less cross-reactivity compared to polyclonal antibodies, which makes them much more effective for therapeutic applications. One of the first approved monoclonal antibodies is OKT3, which reduces acute rejection in organ transplantation patients by blocking the activation of T-cells.
Recombinant antibody production utilizes genetic engineering to create antibodies in vitro by cloning the antibody genes into expression vectors. These vectors are then introduced into host cells, such as bacteria or mammalian cells, to produce the desired antibodies. This method eliminates the need for long-term storage in liquid nitrogen tanks to maintain hybridoma cell lines and provides more consistent results due to minimal batch-to-batch variation.
